Promethazine hydrochloride is a well-known and long-established antihistamine with a variety of other pharmacological effects for
which it is currently used more than for its basic indication of an H1 antihistamine. Since it is, when appropriate, still used mainly in
paediatric patients and since the only dosage form of monocomponent products available in the market is that of a film-coated tablet,
which does not allow for proper splitting when a lower dose is prescribed, it is desirable that the medicinal substance itself be available
for extemporaneous use. It has been available for pharmacies since September 2010. The review article provides information on the
properties, pharmacological effects and the use of promethazine. The aim is not to present the product as a new therapeutic agent
with a high potential in extemporaneous prescription, but to use it for magistral preparation in situations in which no adequate dosage
form of a proprietary medicinal product is available which would allow safe dosage specified in the doctor's prescription. Therefore,
prescriptions intended for extemporaneous preparation of syrups and gelatin capsules are presented.
